Effects of nutrient media, initial seeding rate and shaking speed on the growth of E. coli M15 strain bringing pQE30/PirA recombinant plasmid

 Nội dung tóm tắt: Vibrio has been defined as a cause of Acute Hepatopancreatic Necrosis Disease (AHPND). This disease often occur in Litopenaeus vannamei and Penaeus monodon about 20 - 30 days after stocking, the mortality is able to be up 100% in the severe infected ponds, people have to handle by discharging ponds. AHPND is one of the major concerns of shrimp farming worldwide. The both PirA and PirB genes of Vibrio spp. strains are mainly responsible for causing AHPND in shrimp. In this study, after inserting PirA gene into pQE30 vector and successfully transformed pQE30/PirA recombinant plasmid into E. coli M15 cells. In order to reach an effective expression of PirA gene in E. coli M15 cells, first task is to find out suitable culture conditions for the growth of E. coli M15 strain bringing pQE30/PirA recombinant plasmid. We optimized culture conditions of the E. coli M15 strain including nutrient media, initial seeding rate and shaking speed. The results showed that the E. coli M15 strain grew well in LB medium with initial seeding rate of 4%V and shaking speed of 250 rpm.
